Insulin syringes are graduated in units, not milliliters. A U-100 syringe is built so that 100 units = 1.00 mL, which means one unit = 0.01 mL. That single relationship is the entire conversion. Once you know your peptide concentration in mg/mL, divide by 100 to get mg per unit — or multiply mcg/mL by 0.01 to get mcg per unit.

Two practical notes. First, U-100 barrels come in 0.3 mL (30 unit), 0.5 mL (50 unit), and 1.0 mL (100 unit) sizes — pick the smallest barrel that holds your target volume, because the graduations are finer and the reading error is smaller. Second, a 0.3 mL barrel typically has half-unit markings (0.5 unit = 0.005 mL); a 1.0 mL barrel usually does not. If your target is 12.5 units, the 0.3 mL barrel is the correct tool.

The math, step by step:

  1. Mass of peptide in the vial: e.g. 5 mg.
  2. Volume of diluent added: e.g. 2 mL.
  3. Concentration: 5 mg ÷ 2 mL = 2.5 mg/mL.
  4. Convert to mcg/mL: 2.5 × 1000 = 2500 mcg/mL.
  5. Per unit on U-100: 2500 mcg/mL ÷ 100 units/mL = 25 mcg per unit.

So a 250 mcg target draws to 10 units. A 100 mcg target draws to 4 units. No calculator needed once you have mcg-per-unit.

Read the last two columns carefully. At 10 mg/mL, a 100 mcg draw is one unit — a volume so small that a 1.0 mL barrel cannot resolve it reliably. That is the most common failure mode in this whole workflow: high concentration plus a large barrel equals unreadable graduations. Either dilute further or move to a 0.3 mL barrel.

What syringe should I use for small volumes?

Use the smallest barrel that holds the draw. A 0.3 mL (30 unit) U-100 syringe resolves half-unit increments — 0.005 mL — while a 1.0 mL barrel typically resolves only whole units, 0.01 mL. For any draw under about 20 units, the 0.3 mL barrel cuts your reading error roughly in half.

Three spec-level points worth knowing:

Gauge and length matter less for accuracy than barrel size, but a longer needle adds dead space. For reconstitution work, a fixed-needle 0.3 mL or 0.5 mL U-100 syringe covers nearly every case.

Common conversion errors

The three that show up most:

  1. Using U-40 numbers on a U-100 barrel. A U-40 syringe is 40 units per mL, so one unit = 0.025 mL. Mixing the two scales produces a 2.5× error. Check the barrel cap — it is printed there.
  2. Rounding the concentration before converting. 2.5 mg/mL is 25 mcg/unit exactly. Rounding to "about 2 mg/mL" gives 20 mcg/unit and a 20% error. Keep the concentration exact through the division.
  3. Assuming mg equals units. There is no fixed mg-per-unit value. It is always concentration-dependent. The only fixed number in the chain is 0.01 mL per unit.

A quick sanity check: mcg per unit should always equal mg/mL × 10. At 2.5 mg/mL, that is 25. At 5 mg/mL, 50. If your number does not match that rule, you divided in the wrong direction.
